Amgen Sr Associate Scientist - Assay Development, Implementation & Execution in Thousand Oaks, California
The Therapeutic Discovery organization is seeking a highly motivated and capable Sr Associate Scientist to join the Discovery Technologies department at our Research and Development facility in Thousand Oaks CA (ATO). The successful candidate will be a bench scientist that contributes hands-on to our assay development, implementation, and routine execution efforts in support of Amgen’s small molecule drug discovery pipeline in collaboration with multiple therapeutic areas. The position focuses on the development, optimization, troubleshooting and execution of novel physiologically-relevant cell-based assays supporting HTS, SAR, and MoA studies for promising therapeutic targets. Notably, this individual will design and execute laboratory experiments, generate & interpret data supporting the development of new assays, and test the biological effects of therapeutic agents.
Job responsibilities include:
Develop assays relying on various assay technologies (luminescence, fluorescence, FRET/BRET, High Content Imaging, HTRF, Alpha Screen or similar) implement them in an automated format in 384- or 1536-well plate densities and execute for UHTS, SAR or MOA determination in a timely manner
Apply and incorporate automation solutions to support assay development operations in the lab, assay troubleshooting, mechanism of action and modes of inhibition analysis, data analysis, and reporting
Investigate, identify, develop, and optimize new methods/techniques to promptly generate reliable and reproducible data
Develop sophisticated problem-identification and problem-solving skills
Readily adapt to changing project needs, semi-independently plan, and execute experiments
Maintain fluency with the latest scientific literature and rationale surrounding assay technologies and be an active contributor to the interpretation of results in group discussions
Exhibit excellent teamwork and communication as this role requires an ability to work closely with other scientists in the group and across Amgen
Adhere to good laboratory conduct, documentation practices, and safety guidelines
Doctorate degree OR Doctorate degree completed by October 2019
Master’s degree and 4 years of Scientific experience
Bachelor’s degree and 6 years of Scientific experience
Doctorate or Master's with 3+ years relevant experience working as part of a multi-disciplinary team in a biopharmaceutical setting with a background in Cell and/or Molecular Biology or closely related field
Track record of developing, implementing, and executing novel biochemical or cell-based screening assays in (384 and/or 1536) to support early drug discovery research
Solid background with various assay technologies including luminescence, fluorescence, FRET/BRET, High Content Imaging and with homogenous assays such as HTRF, Alpha Screen or similar
Proficiency with liquid handling equipment (e.g., Echo, Bravo, Tempest, FRD, BioRAPTR, BottleValve, Washer-Dispenser II or similar) and detection instrumentation (e.g., Envision, ViewLux, High Content reader, FLIPR, MSD, or similar), and ability to troubleshoot said instrumentation
Experience with basic molecular biology and biochemistry methods: PCR, qPCR, DNA cloning and protein expression, SDS-PAGE and Western-Blots
Advanced cell culture expertise (immortalized, primary cells or iPS cells), including passaging, transfection, clonal selection, mass production, and cryobanking
Exposure to 3D cell culture systems and ability to grow spheroids
Familiarity with software and statistical platforms for the analysis of large datasets (e.g., Screener, Spotfire, MODDE pro/DoE, Pipeline Pilot)
Previous exposure to large scale automated screening platforms for HTS (GNF, HighRes)
Fluency with High Content Imaging workflows and software (Harmony, Columbus, CellProfiler)
Basic programming skills (Python, R)
A flexible mindset, with strong interpersonal, communication, and problem-solving skills
Demonstrated initiative, teamwork, collaboration skills, an ability to multi-task, and a passion for developing extensive assay development technical expertise
Ability to create and establish priorities, manage work plans, and meet timelines
Capacity to identify, evaluate, implement, and become proficient in novel laboratory techniques
Fluency with GraphPad PRISM, Excel, Word & Powerpoint or similar software
